What is HIC?
The technique known as hydrophobic interaction chromatography (HIC) is a mode of chromatography that separates proteins by differences in surface hydrophobicity. This method utilizes reversible interactions that occur between protein molecules and hydrophobic stationary phase ligands attached to the resin surface.
Certain non-denaturing salts (such as ammonium sulfate or sodium chloride) are used to improve the hydrophobic interactions between proteins and the stationary phase. In most cases, these phases use silica, highly cross-linked agarose, or polystyrene-divinylbenzene as the base particle. Hydrophobic functional groups such as phenyl, ether, octyl (C8), butyl (C4), or amide (among others) are bonded to these resins and interact with the hydrophobic ends of proteins. A reverse gradient is typically run from high-to-low salt concentration in order to effect separation based on relative hydrophobicity.
